Gillingham have been drawn at home to League 2 opposition.
Exeter City, riding high in their own division, will head to Priestfield fort the FA Cup second round match over the weekend of November 28.
Gillingham beat Woking in the first round on Saturday and will be favourites to progress into round three of the FA Cup, when the top teams enter the competition.
Boss Steve Evans reacted to the draw and said: "We wanted a home draw and the balls have been kind.
"Exeter are a team full of good players and a young manager whom I think highly of."
He added: "We will need to produce a good performance to make progress in this country's premier, and distinguished, cup competition."
The Gills at least avoided non-league Brackley Town - twice winners against the Gills in the competition - as they were away to Tranmere Rovers
There were some former Gills players who have taken their teams into round 2, with Mark Bentley’s Canvey Island taking on Boreham Wood and Peter Beadle’s Barnet home to MK Dons.
Exeter, who sit fourth in League 2, overcame National League North side AFC Fylde at the weekend, winning the game 2-1.
The FA Cup match will be the first meeting between Exeter and Gillingham at Priestfield for eight years, the visitors winning that game in November 2012 3-2.
The last meeting between the teams in the FA Cup came in 1935, the Gills winning that game 4-0. Forward Bill Baldwin got a hat-trick in the win.
