Joe Taylor scored both goals as Margate recorded their first home league win since the opening day of the Bostik Premier season.
Taylor's 21st minute penalty was cancelled out before half-time by Billy Bricknell, but the Gate's former Folkestone striker had the final say, popping up with a 75th minute winner.
Taylor's goals were Margate's first in the league at Hartsdown since they beat Corinthian Casuals 5-2 on the first Saturday.
The victory was their first in five before their own supporters and signalled the end of Enfield's unbeaten record.
Tom Wynter returned to the Gate starting line-up, replacing right-back Jack Paxman who dropped to the bench, and the home side made a bright start.
They forced two early corners and kept Enfield pinned in their own half for much of the opening 20 minutes.
Their best moment during that period came on 15 minutes when Taylor produced a nice touch and lay-off to George Brown but his team-mate was unable to pick him out in the box with his return pass.
However six minutes later the two men were to feature prominently in the game's opening goal as Brown was tripped in the box by Enfield's Aaron Greene and Taylor sent keeper Joe Wright the wrong way from the spot.
Soon after Sam Youngs shot wide from Enfield's first real opening but they hadn't really threatened an equaliser when former Dover striker Bricknell picked the ball up on the Gate right and turned to hit a precise finish into the far corner.
The goal prompted their best spell of the half with three corners in five minutes and during the opening minutes of the second half it looked like the tide had started to turn.
Midfielder Youngs was becoming increasingly influential and he created the first chance of the second half when he was allowed to run from deep before hitting a low shot which home keeper Henry Newcombe managed to push wide.
Moments later Gate skipper Liam Friend produced a terrific block at the near post and then, from a short free-kick Bricknell's low drive hit the foot of the Gate post.
Bricknell also fired wide from the edge of the box while at the other end, Gate centre-half Ben Swift, up for a free-kick, lofted over from an acrobatic overhead kick.
Gate came back on 68 minutes when Frannie Collin's corner was met by Wynter whose header was turned aside by Wright.
From the next corner there were unsuccessful appeals for a penalty from the home side when Swift went down under a challenge from Taofiq Olomowewe.
The big Enfield defender then popped up at the other end when he headed wide from eight yards out when he should have scored and within 90 seconds Margate had made him pay.
Taylor was the hero again, wriggling free in the box to chest the ball down before nonchalantly lofting over Wright for his seventh goal of the campaign.
Newcombe preserved the lead by getting down smartly to deny Youngs soon after but thereafter the Gate stood firm and there were fervent appeals for another penalty late on when substitute Alex Flisher looked to have been fouled by Enfield skipper Ryan Blackman.
