Tunbridge Wells Borough Council have joined a list of authorities opposed to expansion at Gatwick airport.
The local council voted on the issue at a meeting on Wednesday night with 40 votes against expansion, one in favour and one abstention.
The decision followed a public debate in which councillors heard from aviation experts and local people.
No representatives from Gatwick Airport were at the meeting.
The vote against the prospect of a second runway is the latest in a series of setbacks for the proposed development with a decision due from The Airports Commission after this year's General Election.
